TVR Speed 12 is not eligible; i'm not sure about the Gillet and Stealth either. I'm sure about the TVR though since only one road car was ever made.

Would the Aston Martin Vanquish (or DB7 GT) be eligible?

I'm guessing that neither are eligible for GTS- although a Stealth DID run in Grand-Am GTS a couple of times last year,- although it was renamed 'Predator' possibly for reasons to do with licensing of brand names in the USA (think the 'Stealth' name is used by one of the US motor manufacturers?)

Does it need to be a mass production car (more than 100 cars built)?

If so, then the Gillet Vertigo is certainly not eligible since there are only a handfull of racecars made.

Evil Fuzzy should be qualifying in the top 10 on the grid
The Gillet Vertigo isn't eligible. Not only because of the number of cars built, but because the race version is a GT class car (it races in the GTB class of the Belcar against Porsches GT3-R(S), Marcos Mantis etc.)
